When creating an assessment, is it possible to draw a set number of questions from a random question pool? I.e 20 random questions from a pool of 200 questions.

Unfortunately, this isn't something you can do with the current version of the contrib-assessment plugin. It is however a feature we're working hard to get into the new 2.0 release of the framework and core bundle.
It might be worth having a look at Kineo's fork of the assessment, as this has banking and randomisation (although beware that this may rely on other custom code not included in the plugin).
